Naldemedine Tosylate falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Ayahuasca interacts with.

Theoretically, ayahuasca might increase plasma concentrations of CYP3A4 substrates.
Ayahuasca contains beta-carboline alkaloids. In vitro research suggests that beta-carboline alkaloids competitively inhibit CYP3A4 enzymes. This effect has not been reported in humans.

Here's the gist for your Symproic (naldemedine). Your body relies on a liver enzyme called CYP3A4 to clear this medicine out. The beta-carboline compounds in ayahuasca can slow that same enzyme down, at least in lab (test tube) studies. If that happened in a person, naldemedine could build up a bit higher than usual, and you might notice more of its typical effects, like stomach cramps, diarrhea, belly pain, or nausea.

I want to be honest about the strength here: this is theoretical and based only on lab research, not real-world human cases, so it may not amount to much. That said, ayahuasca carries its own serious risks. Please don't start, stop, or adjust anything without checking with your doctor or pharmacist first.

What Is Naldemedine Used For?

Brand name: Symproic

Symproic (naldemedine) is used to treat opioid-induced constipation (OIC) — constipation caused by opioid pain medications — in adults with chronic non-cancer pain. This includes people living with chronic pain related to prior cancer or its treatment, as long as they do not need frequent opioid dose increases (such as weekly adjustments).

It is not a general constipation remedy. Symproic works specifically against the constipating effect of opioids and should be stopped if you stop taking your opioid pain medication.
