Ponesimod falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Ayahuasca interacts with.

Theoretically, ayahuasca might increase plasma concentrations of CYP3A4 substrates.
Ayahuasca contains beta-carboline alkaloids. In vitro research suggests that beta-carboline alkaloids competitively inhibit CYP3A4 enzymes. This effect has not been reported in humans.

Here's the gist for Ponvory (ponesimod). Your body relies partly on an enzyme called CYP3A4 to clear this drug. The beta-carboline compounds in ayahuasca have shown, in lab dish studies only, that they can block CYP3A4. If that blocking happened in real life, ponesimod could clear more slowly and build up, which might mean stronger effects and more side effects like a slowed heart rate (especially when you first start the medicine), shortness of breath, swelling, headache, or higher blood pressure.

I want to be honest though: this is theoretical. It has not actually been seen in people, so it's a maybe, not a sure thing. Ayahuasca carries its own serious risks too. Please don't change anything on your own, just loop in your doctor or pharmacist first.
