Here's the simple version: amphetamine sulfate is already a strong stimulant. Bitter orange supplements contain synephrine, which acts a lot like adrenaline and is itself a mild stimulant. When you stack two stimulants together, their effects can add up. With amphetamine, that means a bigger chance of a racing or pounding heart, higher blood pressure, jitteriness, anxiety, and trouble sleeping.

This isn't a metabolism problem where one blocks the other. It's more like pressing the gas pedal twice at once. The concern is mostly theoretical and based on limited evidence, but it makes sense given how both work. If you take amphetamine for ADHD or another reason, please check with your pharmacist or doctor before adding any bitter orange or weight-loss/energy product, especially if you have any heart or blood pressure issues.

Theoretically, bitter orange might increase levels of drug metabolized by CYP2D6.
In vitro research shows that octopamine, a constituent of bitter orange, weakly inhibits CYP2D6 enzymes. This effect has not been reported in humans.

Here's the situation with bitter orange and your amphetamine. Part of how your body clears amphetamine runs through a liver enzyme called CYP2D6. In lab dishes, a compound in bitter orange (octopamine) weakly slowed that enzyme down. If that happened in real life, your amphetamine could clear a little slower and build up slightly, which might nudge up the usual effects: faster heartbeat, higher blood pressure, jitteriness, trouble sleeping, or feeling wired.

The bigger concern, honestly, is that both bitter orange and amphetamine are stimulants. Stacking them can add together and push your heart rate and blood pressure higher, even apart from the enzyme question. The enzyme part is only theoretical (test-tube data, never shown in people), so I wouldn't panic, but please check with your pharmacist or doctor before combining them.
