Entrectinib falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Dhea interacts with.

Theoretically, DHEA might increase the levels of drugs metabolized by CYP3A4.
Some preliminary evidence shows that DHEA may inhibit CYP3A4; however, the clinical significance of this potential interaction is not known.

Here's the gist with Rozlytrek (entrectinib) and DHEA. Your liver uses an enzyme called CYP3A4 to clear entrectinib out of your system. There's some early, anecdotal evidence that DHEA may slow that enzyme down a bit. If that happens, entrectinib could build up a little higher than expected, which can mean more of its usual side effects, things like dizziness, fatigue, swelling, constipation, changes in taste, or effects on your heart rhythm.

I want to be honest that this is theoretical and weakly supported, so we don't really know if it matters in real life. But entrectinib is a serious cancer drug, so please don't start, stop, or combine anything without first checking with your oncologist or pharmacist.

What Is Entrectinib Used For?

Brand name: Rozlytrek

Rozlytrek is used to treat adult patients with ROS1-positive metastatic non-small cell lung cancer (NSCLC) — a type of lung cancer where the ROS1 gene is rearranged — as confirmed by an FDA-approved test.

Rozlytrek is also approved for adult and pediatric patients older than 1 month with solid tumors that have an NTRK gene fusion (a specific genetic alteration), detected by an FDA-approved test, that have spread or cannot be removed without causing serious harm, and have worsened after prior treatment or have no good treatment alternatives. This NTRK indication was granted accelerated approval based on tumor response rates; continued approval may depend on results from confirmatory trials.
