Amitriptyline, Chlordiazepoxide falls under CYTOCHROME P450 2C9 (CYP2C9) SUBSTRATES — the group Diosmin interacts with.

Theoretically, diosmin might inhibit the metabolism of CYP2C9 substrates.
Diclofenac is metabolized by CYP2C9 enzymes. Clinical and laboratory research shows that diosmin inhibits the metabolism of diclofenac. A pharmacokinetic study in humans shows that taking diosmin (Venex 500) 500 mg daily for 9 days prior to oral administration of diclofenac 100 mg increases blood levels of diclofenac and decreases diclofenac clearance.

Here's the gist for Limbitrol, which combines amitriptyline (an older antidepressant) with chlordiazepoxide (a benzodiazepine). Diosmin may gently slow down a liver enzyme called CYP2C9. In theory, that could let a little more of these medicines build up in your body rather than clearing out at the usual pace. For you, that might mean feeling extra drowsy, lightheaded, or dry-mouthed, with possible constipation, blurry vision, or grogginess the next morning.

I want to be honest though: this concern is theoretical, based on how the enzyme works and a study using a different drug (diclofenac), not on real-world reports with Limbitrol. Both ingredients also rely on several other enzymes, so the effect may be small. Still, since these meds already cause sedation, it's worth a quick chat with your pharmacist or doctor before combining them.

Theoretically, diosmin might inhibit the metabolism of CYP3A4 substrates.
Laboratory research is conflicting with respect to the effects of diosmin on CYP3A4. Some research suggests that diosmin does not affect CYP3A4 activity. However, other research suggests that diosmin alters the metabolism of carbamazepine, a CYP3A4 substrate. Laboratory and animal research show that oral administration of diosmin for 7 days prior to oral administration of carbamazepine increases plasma concentrations of carbamazepine, decreases the clearance of carbamazepine, and decreases the formation of carbamazepine's active metabolite. Additionally, pharmacokinetic research in healthy male subjects shows that taking diosmin (Venex) 500 mg daily for 10 days prior to oral administration of carbamazepine 200 mg increases blood levels of carbamazepine by approximately 58% and decreases carbamazepine clearance by 42%. It is speculated that diosmin reduces the metabolism of carbamazepine by inhibiting CYP3A4. Diosmetin, a metabolite of diosmin, may also inhibit CYP3A4.

Limbitrol DS combines amitriptyline (an older antidepressant) with chlordiazepoxide (a benzodiazepine for anxiety). Both rely partly on a liver enzyme called CYP3A4 to be broken down. The idea here is that diosmin may slow that enzyme down a bit, so these medicines could clear out of your body more slowly and build up to slightly higher levels.

If that happens, you might notice more of their usual effects, such as drowsiness, dizziness, dry mouth, constipation, or feeling foggy or unsteady. I want to be honest, though: this is mostly theoretical, the lab data is mixed, and the human study was done with a different drug (carbamazepine). It's not a sure thing. Still, since both ingredients can already cause sedation, it's worth a quick chat with your pharmacist or doctor before adding diosmin, especially if you feel extra sleepy.
