Lesinurad falls under CYTOCHROME P450 2C9 (CYP2C9) SUBSTRATES — the group Diosmin interacts with.

Theoretically, diosmin might inhibit the metabolism of CYP2C9 substrates.
Diclofenac is metabolized by CYP2C9 enzymes. Clinical and laboratory research shows that diosmin inhibits the metabolism of diclofenac. A pharmacokinetic study in humans shows that taking diosmin (Venex 500) 500 mg daily for 9 days prior to oral administration of diclofenac 100 mg increases blood levels of diclofenac and decreases diclofenac clearance.

Here's the gist: your body clears lesinurad partly through a liver enzyme called CYP2C9. Diosmin can gently slow that same enzyme down. So in theory, pairing the two could let lesinurad linger a bit longer in your system, which may nudge up its effects and side effects. With lesinurad, the things to keep an eye on are kidney-related, since it works by pushing uric acid out through the kidneys. Headache, reflux, or signs of kidney strain are worth noting.

Now, the honest part: this is a theoretical concern based on how the enzyme behaves and on a study using a different drug (diclofenac), not lesinurad itself. It may never cause you any trouble. Still, lesinurad already carries kidney cautions, so I'd run this combo by your pharmacist or doctor before starting diosmin.

Diosmin is a citrus-derived flavonoid most often used for vein-related problems such as chronic venous insufficiency and hemorrhoids, and it is usually combined with a related flavonoid called hesperidin. Some studies suggest it may help relieve leg swelling and hemorrhoid symptoms, but quality varies. What Is Lesinurad Used For?

  • Lesinurad is used in combination with a xanthine oxidase inhibitor to treat hyperuricemia (high levels of uric acid) in people with gout (sudden attacks of redness, swelling, pain, and heat in one or more joints) whose disease is not controlled with their current medication. Lesinurad is in a class of medications called selective uric acid reabsorption inhibitors. It works by helping the kidneys to remove uric acid from the body.
