Adagrasib falls under HEPATOTOXIC DRUGS — the group Padang Cassia interacts with.

There is some concern that ingesting large amounts of Padang cassia might cause hepatotoxicity in some people. Padang cassia contains coumarin. Coumarin has caused hepatotoxicity in animal models. In otherwise healthy humans, very high doses of coumarin, from 50-7000 mg daily, can result in hepatotoxicity that resolves when coumarin use is discontinued. Lower amounts might also cause liver problems in sensitive people, such as those with liver disease or those taking potentially hepatotoxic agents.

Here's the thing with Krazati (adagrasib): it's a targeted cancer pill that can be tough on the liver on its own. Many people taking it already get raised liver enzymes, and their care team keeps a close eye on bloodwork during treatment. So the worry here isn't that cinnamon changes how adagrasib works, it's that piling two liver-stressing things together could add up.

Padang cassia contains coumarin, which in large, concentrated supplement doses has caused liver trouble in some people. This concern is mostly theoretical, based on how coumarin behaves, not on studies of these two together. Sprinkling cinnamon on your food is not a problem. But before taking concentrated cinnamon capsules with adagrasib, check with your oncology pharmacist or doctor first.

Adagrasib also falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES.

In vitro research shows that a methanol extract of Padang cassia inhibits cytochrome P450 3A4 (CYP3A4) activity. Theoretically, taking Padang cassia with CYP3A4 substrates might increase the effects and side effects of these substrates.

Adagrasib (Krazati) is a targeted cancer pill used for certain lung cancers, and your body relies on a liver enzyme called CYP3A4 to break it down. Lab studies suggest concentrated Padang cassia cinnamon may slow that same enzyme down a bit. In theory, if the enzyme works slower, adagrasib could build up in your blood and lead to stronger side effects, such as nausea, diarrhea, fatigue, or changes in liver and heart (QT) lab tests.

Keep in mind this is only a test-tube finding, not something proven in people, so I wouldn't lose sleep over it. Sprinkling cinnamon on your food is very unlikely to matter. Daily high-dose cinnamon capsules are a different story, so please run those by your oncology team or pharmacist first.

Padang Cassia is a type of cinnamon (Indonesian or Korintje cinnamon) grown mainly in Indonesia and used widely as a kitchen spice. As a supplement, evidence for health benefits like blood sugar control is limited and mixed, and like other cassia cinnamons it naturally contains coumarin, which can be harmful to the liver in large amounts. Use food amounts freely, but talk to a pharmacist or doctor before taking concentrated cinnamon supplements.

About Adagrasib

What Is Adagrasib Used For?

Brand name: Krazati

Krazati (adagrasib) is used to treat adults with locally advanced or metastatic non-small cell lung cancer (NSCLC) that carries a specific genetic change called a KRAS G12C mutation. It is used as a single agent for NSCLC after at least one prior treatment, and patients must be confirmed to have this mutation by an FDA-approved test.

Krazati is also used in combination with another drug called cetuximab to treat adults with KRAS G12C-mutated locally advanced or metastatic colorectal cancer (CRC) who have already been treated with specific prior chemotherapy regimens. Both uses were approved under the FDA's accelerated approval program based on tumor response data, and continued approval may depend on results from confirmatory trials.
