Letrozole falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Padang Cassia interacts with.

In vitro research shows that a methanol extract of Padang cassia inhibits cytochrome P450 3A4 (CYP3A4) activity. Theoretically, taking Padang cassia with CYP3A4 substrates might increase the effects and side effects of these substrates.

Here's the simple version: your body clears letrozole partly through a liver enzyme called CYP3A4. In a test-tube study, a concentrated extract of Padang cassia slowed that enzyme down a bit. If that held true in people, letrozole could clear more slowly and build up slightly, which in theory might nudge up its usual effects, things like joint or muscle aches, hot flashes, tiredness, or headache.

I want to be honest about the strength of this, though. It's based only on lab work, not on real patients, so it's quite theoretical. Sprinkling cinnamon on your oatmeal is very unlikely to matter. The question mark is really about concentrated cinnamon capsules taken daily. Since letrozole is an important cancer medicine, just run any cinnamon supplement past your oncologist or pharmacist first.

Padang Cassia is a type of cinnamon (Indonesian or Korintje cinnamon) grown mainly in Indonesia and used widely as a kitchen spice. As a supplement, evidence for health benefits like blood sugar control is limited and mixed, and like other cassia cinnamons it naturally contains coumarin, which can be harmful to the liver in large amounts. Use food amounts freely, but talk to a pharmacist or doctor before taking concentrated cinnamon supplements.

What Is Letrozole Used For?

Brand name: Femara

Letrozole (brand name Femara, or available as generic letrozole tablets) is used to treat breast cancer in postmenopausal women whose tumors are hormone receptor positive — meaning the cancer feeds on estrogen. It is used in several situations: as an adjuvant treatment (therapy given after primary treatment to reduce the risk of the cancer returning) for early breast cancer, and as extended adjuvant treatment for women who have already completed five years of tamoxifen therapy.

Femara and letrozole tablets are also used as first-line or second-line treatment for locally advanced or metastatic (spread to other areas) breast cancer in postmenopausal women, including cases where the cancer has progressed after other anti-estrogen therapy.
