This combo pill pairs amitriptyline (a tricyclic antidepressant) with perphenazine (an antipsychotic). Both of these medicines are already known to lower the seizure threshold a bit, meaning they can make seizures slightly easier to trigger in people who are prone to them.

The concern here is mostly theoretical. In animal lab research, rauwolscine lowered the seizure threshold too, so stacking it on top of medicines that already do this might add up and raise the chance of a seizure. We don't have human studies proving this, so I wouldn't panic. Still, if you have any history of seizures or take this combination, it's worth a quick chat with your doctor or pharmacist before adding a stimulant supplement like this one.

Theoretically, rauwolscine might increase levels of drugs metabolized by CYP2D6.
Rauwolscine is structurally related to yohimbine. In vitro research shows that yohimbine inhibits CYP2D6 enzyme activity.

Triavil combines amitriptyline (an older antidepressant) with perphenazine (an antipsychotic), and both of these rely on a liver enzyme called CYP2D6 to be cleared from your body. Rauwolscine is a cousin of yohimbine, and in lab studies yohimbine appears to slow down that same enzyme. So in theory, pairing them could let these two drugs build up a bit higher than usual.

If that happened, you might notice more of their typical side effects: drowsiness, dry mouth, constipation, dizziness when standing, blurred vision, or a faster heartbeat. Keep in mind this is theoretical, based only on test-tube data, not real-world reports. Still, since rauwolscine itself raises heart rate and blood pressure, it's worth running this combo past your pharmacist or doctor first.

Rauwolscine (alpha-yohimbine) is a stimulant alkaloid closely related to yohimbine that is marketed mainly in fat-burning and pre-workout supplements. Human evidence for its benefits is very limited, and it can raise heart rate and blood pressure and cause anxiety, so it should be used with caution and is not recommended for people with heart conditions, anxiety, or during pregnancy or breastfeeding.
