Rauwolscine and Amphetamine Sulfate

These two interact in 3 distinct ways. Each one is explained separately below.

Why Are There 3 Interactions?

Rauwolscine interacts with whole categories of medication. Amphetamine Sulfate falls into 3 of those categories, so it can interact with Rauwolscine in 3 different ways. We’ve listed each one below — your overall risk is driven by the most serious of them (Moderate).

Moderate

SEIZURE THRESHOLD LOWERING DRUGS

1 of 3

Why this applies to Amphetamine Sulfate

Amphetamine Sulfate falls under SEIZURE THRESHOLD LOWERING DRUGS — the group Rauwolscine interacts with.

Theoretically, taking rauwolscine with seizure threshold lowering drugs might increase the risk of adverse convulsant effects.
In animal research, intraperitoneal rauwolscine lowered the seizure threshold level of the drug metrazol.
Severity Moderate Likelihood Possible Evidence D

Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.

Here's the simple version: both rauwolscine and amphetamine are stimulants that rev up your nervous system. Amphetamine already pushes out norepinephrine and dopamine to keep you alert, and rauwolscine adds even more stimulant signaling by blocking the brakes (alpha-2 receptors). Stacking two stimulants together can mean a faster heart rate, higher blood pressure, jitteriness, anxiety, and trouble sleeping. The specific worry on file is that rauwolscine may lower the seizure threshold, which could nudge up the small risk of a seizure.

To be fair, this seizure concern comes only from animal lab research, so it's theoretical and not proven in people. Still, since you'd be combining two stimulants, it's worth running this by your pharmacist or doctor before mixing them.

CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES

2 of 3

Amphetamine Sulfate also falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES.

Theoretically, rauwolscine might increase levels of drugs metabolized by CYP2D6.
Rauwolscine is structurally related to yohimbine. In vitro research shows that yohimbine inhibits CYP2D6 enzyme activity.
Severity Moderate Likelihood Possible Evidence D

Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.

Dr. Brian Staiger, PharmD, BCPS

Here's the thing to keep in mind if you pair these two. Amphetamine sulfate (Benzedrine, Evekeo ODT) is partly cleared by a liver enzyme called CYP2D6. Rauwolscine is a cousin of yohimbine, and lab studies suggest yohimbine can slow that enzyme down. If rauwolscine does the same, your body might clear amphetamine a bit more slowly, so more of it could hang around in your system.

On its own, that could mean stronger and longer stimulant effects: a faster or pounding heartbeat, higher blood pressure, jitteriness, trouble sleeping, and more anxiety. And remember, rauwolscine is itself a stimulant, so you'd be stacking two things that rev up the body. This is a theoretical concern based on lab data, not proven in people, but I'd still run it by your pharmacist or doctor before combining them.

STIMULANT DRUGS

3 of 3

Amphetamine Sulfate also falls under STIMULANT DRUGS.

Theoretically, taking rauwolscine with stimulant drugs might increase the risk of adverse stimulant effects.
Rauwolscine has demonstrated stimulant effects in animal research.

Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.

Dr. Brian Staiger, PharmD, BCPS
Dr. Brian Staiger explains PharmD, Clinical Pharmacist

Here's the simple version. Amphetamine sulfate already revs up your body by boosting norepinephrine and other stimulant chemicals. Rauwolscine works through a different door, blocking alpha-2 receptors, but it pushes in the same direction by raising those same stimulant signals. So stacking the two could add up and make stimulant-type side effects more likely, things like a faster heart rate, higher blood pressure, jitteriness, trouble sleeping, and anxiety.

I want to be honest about the evidence, though. This concern comes mostly from lab and animal studies, so it's more of a theoretical caution than a proven problem in people. Still, two stimulants together is worth a real conversation. Please check with your pharmacist or doctor before combining them, especially if you have any heart or anxiety issues.

Major

Clinically significant. Avoid the combination, or use only under close professional supervision.

Moderate

Usually worth avoiding or managing — may need monitoring, a dose change, or timing apart.

Minor

Generally not serious. Be aware of it and mention it to your pharmacist.

Likelihood how strong the evidence is that the interaction actually happens in people
  • LikelyWell-controlled human studies have demonstrated the likely existence of this interaction.
  • ProbableInteraction has not been documented in well-controlled studies, however, the interaction has been demonstrated in some small human studies or in controlled animal studies in conjunction with multiple case reports.

From our pharmacist’s overview on the Amphetamine Sulfate page

“Amphetamine sulfate (Benzedrine, Evekeo ODT) is a prescription stimulant, and most of what earns this list genuine respect is more stimulation piled on top of it. What is Rauwolscine?

  • Fat loss and weight management
  • Bodybuilding and physique support
  • Energy and stimulant effects
  • Athletic performance
  • Libido support

Rauwolscine (alpha-yohimbine) is a stimulant alkaloid closely related to yohimbine that is marketed mainly in fat-burning and pre-workout supplements. Human evidence for its benefits is very limited, and it can raise heart rate and blood pressure and cause anxiety, so it should be used with caution and is not recommended for people with heart conditions, anxiety, or during pregnancy or breastfeeding.
