CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES

Eszopiclone falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Rhodiola interacts with.
In vitro research shows that rhodiola inhibits CYP3A4. This effect is highly variable and appears to be dependent on the rhodiola product studied. However, a clinical study in healthy young males found that taking rhodiola extract 290 mg daily for 14 days does not inhibit the metabolism of midazolam, a CYP3A4 substrate.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the gist: your body clears eszopiclone (Lunesta) using a liver enzyme called CYP3A4. In test-tube studies, Rhodiola has shown some ability to slow that same enzyme down. If that were to happen in real life, eszopiclone could build up a bit and hit harder than usual, meaning more next-day drowsiness, grogginess, dizziness, or trouble concentrating in the morning.
That said, I'd keep this in perspective. The concern comes mostly from lab data, and a human study actually found Rhodiola did not meaningfully slow CYP3A4 for a similar drug. So this is rated minor and theoretical. Still, since both can make you sleepy, it's worth a quick chat with your pharmacist or doctor before pairing them.

A review of adverse event reports in Poland identified cases of excessive sedation, myoclonus, hypotension, and hallucinations when rhodiola was taken with haloperidol, diazepam, or alprazolam.

If you take Lunesta (eszopiclone) to help you sleep, here's the gist with Rhodiola: this is a fairly minor and mostly theoretical concern, but it's worth knowing. Eszopiclone already calms brain activity to bring on sleep. Rhodiola may have a mild calming or sedating effect of its own, so stacking the two could, in theory, leave you feeling more drowsy than expected, groggy or unsteady the next morning, or a bit lightheaded.
The evidence here is weak, based mostly on a handful of anecdotal reports with other sedatives, not eszopiclone specifically. Eszopiclone isn't a prodrug, so this isn't about one speeding up or blocking the other. Most people do fine, but if you notice extra grogginess or dizziness, please check in with your pharmacist or doctor before changing anything.

Brand name: Lunesta
Eszopiclone tablets — sold under the brand name Lunesta and as generics — are indicated for the treatment of insomnia. In clinical studies, eszopiclone helped people fall asleep more quickly (reduced sleep latency) and stay asleep through the night (improved sleep maintenance).
Clinical trials supporting its effectiveness lasted up to 6 months in adults and up to 2 weeks in elderly patients. Lunesta and eszopiclone generics are approved for this same indication across the available tablet formulations.
