Darunavir falls under CYTOCHROME P450 1A2 (CYP1A2) INDUCERS — the group Sassafras interacts with.

In vitro research suggests that cytochrome P450 1A2 (CYP1A2) inducers might increase the cytotoxicity of safrole. So far, this interaction has not been reported in humans. Also, it is not known if CYP1A2 inducers increase the toxicity of sassafras. Inducers of CYP1A2 include omeprazole (Prilosec).

Here's the gist for someone taking Darunavir (Prezista) alongside sassafras. The concern isn't that sassafras changes your HIV medicine. It's the other way around. Safrole, the main compound in sassafras, can be turned into more harmful, liver-damaging byproducts by a liver enzyme called CYP1A2. Drugs that rev up that enzyme may make safrole more toxic. This idea comes only from lab and animal studies, and it hasn't been seen in real people, so think of it as theoretical for now.

Honestly, the bigger point is that sassafras itself contains safrole, which the FDA banned in foods over cancer and liver concerns. I'd skip internal use entirely and pick safrole-free products. Please run any supplement past your pharmacist or doctor before mixing it with your HIV regimen.

Sassafras is an aromatic North American tree whose root bark and oil were once popular in teas and as a flavoring, but it contains safrole, a compound the FDA has banned in foods because it can cause cancer and liver damage. There is no good evidence it safely treats any health condition, and most experts recommend avoiding internal use. If you want the flavor, choose products that are labeled safrole-free.

About Darunavir

What Is Darunavir Used For?

Brand name: Prezista

Darunavir treats HIV-1 infection in adults and in children who are at least 3 years old and weigh at least 10 kg (about 22 lbs). It is not a cure for HIV, but it is used as part of a combination antiretroviral regimen to control the virus.

Darunavir must always be taken together with ritonavir — that pairing is required for darunavir to work properly. Both Prezista (brand name) and generic darunavir tablets are approved for this use. Prezista is also available as an oral suspension, which is particularly useful for younger or smaller patients.
