Topiramate falls under CNS DEPRESSANTS — the group Sassafras interacts with.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the simple version: sassafras may have mild sedating, calming effects of its own. Topiramate (Topamax) can already make people feel drowsy, foggy, dizzy, or slowed down in their thinking. So the worry is that putting the two together could stack those effects and leave you more tired or out of it than usual. Topiramate isn't a prodrug here, so this isn't about one activating the other, it's just an additive sleepiness concern.
I'll be honest, this interaction is theoretical, based on how sassafras behaves, not on real patient reports. The bigger issue is that sassafras contains safrole, which the FDA banned in foods because of liver and cancer risks, so I'd steer clear of it anyway. Please check with your pharmacist or doctor before combining them.

Sassafras is an aromatic North American tree whose root bark and oil were once popular in teas and as a flavoring, but it contains safrole, a compound the FDA has banned in foods because it can cause cancer and liver damage. There is no good evidence it safely treats any health condition, and most experts recommend avoiding internal use. If you want the flavor, choose products that are labeled safrole-free.

Topiramate is used alone or with other medications to treat certain types of seizures including primary generalized tonic-clonic seizures (formerly known as a grand mal seizure; seizure that involves the entire body) and partial onset seizures (seizures that involve only one part of the brain). Topiramate is also used with other medications to control seizures in people who have Lennox-Gastaut syndrome (a disorder that causes seizures and developmental delays). Topiramate is also used to prevent migraine headaches but not to relieve the pain of migraine headaches when they occur. Topiramate is in a class of medications called anticonvulsants. It works by decreasing abnormal excitement in the brain.
