In vitro research shows that triptolide, a constituent of thunder god vine, inhibits CYP1A2 enzymes in human microsomes. This interaction has not been reported in humans.

Here's the gist for Zileuton. Your body relies on a liver enzyme called CYP1A2 to help clear this asthma medicine. In lab dish studies, a compound in thunder god vine (triptolide) seems to slow that same enzyme down. If that happened in people, Zileuton could build up a bit higher than expected, which might mean more of its usual side effects, things like nausea, stomach upset, headache, or stress on the liver (Zileuton already requires liver enzyme monitoring).

I'll be honest with you though: this is theoretical. It's based only on test-tube work and has never actually been reported in a real person. Still, since thunder god vine itself can be toxic if not used carefully, please loop in your doctor or pharmacist before combining the two.

Theoretically, CYP3A4 inhibitors might increase the levels and adverse effects of thunder god vine constituents.
Inhibitors of CYP3A4 can increase the concentration of triptolide, a thunder god vine constituent, and its derivative (5R)-5-hydroxytriptolide. In vitro research shows that triptolide and (5R)-5-hydroxytriptolide are about 94% and 64% metabolized by CYP3A4, respectively.

Zileuton can slow down a liver enzyme called CYP3A4, and that same enzyme is what your body uses to break down the active compounds in Thunder God Vine (like triptolide). So when Zileuton blocks that pathway, those herbal compounds may clear more slowly and build up to higher levels than expected.

Why that matters: Thunder God Vine can already be hard on the body, with possible stomach upset, diarrhea, and at higher amounts more serious toxicity. More of it hanging around could mean more of those effects. I'll be honest, this is a theoretical concern based on how the drug and herb behave, not proven cases. What Is Zileuton Used For?

Brand names: Zyflo, Zyflo CR

Zileuton is used for the ongoing prevention and treatment of asthma in adults and adolescents aged 12 and older. It is a maintenance medicine — meaning it is taken regularly to keep asthma under control, not to stop an asthma attack once it has started.

Zyflo (the tablet form) and zileuton extended-release tablets are both approved for this same purpose. Neither form should be used to treat a sudden asthma attack. However, both can be continued during a flare-up of asthma while you also use your rescue inhaler.
