ADO-Trastuzumab Emtansine falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Tree Turmeric interacts with.

Theoretically, tree turmeric might increase the levels and clinical effects of drugs that are substrates of CYP3A4.
In vitro research and preliminary clinical evidence suggest that berberine, a constituent of tree turmeric, moderately inhibits CYP3A4.

Here's the gist for Kadcyla (ado-trastuzumab emtansine). Once this antibody-drug conjugate releases its cancer-killing payload (a chemo molecule called DM1), that piece is cleared with help from a liver enzyme named CYP3A4. The berberine in tree turmeric can mildly slow that enzyme down, so in theory the toxic payload could linger a bit longer and hit harder.

What that might look like in real life is more of Kadcyla's known side effects, things like low platelets, easy bruising or bleeding, liver enzyme changes, tiredness, or nerve tingling. I'll be honest though, this is mostly a theoretical, lab-based concern, not something proven in people on Kadcyla. Since this is serious cancer therapy, please don't add tree turmeric without first clearing it with your oncology team or pharmacist.

About Ado-trastuzumab Emtansine Injection

What Is Ado-trastuzumab Emtansine Injection Used For?

Brand name: Kadcyla

Kadcyla (ado-trastuzumab emtansine) is used on its own to treat HER2-positive metastatic breast cancer in adults who have already received trastuzumab and a taxane chemotherapy, either together or separately. It is used when cancer has come back or spread, or when the disease returned within six months of finishing earlier treatment.

Kadcyla is also used as adjuvant (after-surgery) treatment for HER2-positive early breast cancer in patients who still had remaining cancer after completing neoadjuvant (pre-surgery) treatment with a taxane and trastuzumab. Patients must be tested for HER2 overexpression or gene amplification using an FDA-authorized test before starting Kadcyla.
