Mitapivat falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Yerba Mate interacts with.

Theoretically, yerba mate might increase the levels and clinical effects of CYP3A4 substrates.
In vitro research shows that yerba mate extract inhibits CYP3A4 enzymes. Theoretically, taking yerba mate may increase levels and adverse effects of CYP3A4 substrates.

Here's the quick version: your body clears mitapivat (Pyrukynd) using a liver enzyme called CYP3A4. In a test tube, yerba mate appears to slow that enzyme down a bit. In plain terms, if mitapivat is broken down more slowly, a little more could linger in your system, which in theory might nudge up side effects like headache, nausea, back pain, or trouble sleeping.

I want to be honest about how soft this is. The concern comes only from lab studies, not from people actually taking the two together, so it's labeled minor and theoretical. A casual mug of mate is unlikely to be a problem, while strong concentrated extracts are the bigger question mark. Just give your pharmacist or doctor a heads-up so they can keep an eye on you.

About Mitapivat

What Is Mitapivat Used For?

Brand name: Pyrukynd

Mitapivat is available as two separate branded products approved for different conditions. Pyrukynd is used to treat hemolytic anemia — a type of anemia caused by red blood cells breaking down too quickly — in adults who have pyruvate kinase (PK) deficiency, a rare inherited blood disorder. Aqvesme is used to treat anemia in adults with alpha- or beta-thalassemia, inherited conditions that affect how the body makes hemoglobin.

These are distinct approvals. Pyrukynd and Aqvesme are not interchangeable, and each is approved only for its specific indication.
