Why this applies to Aspirin, Caffeine

Aspirin, Caffeine falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Aegeline interacts with.

Aegeline inhibits cytochrome P450 3A4 (CYP3A4) activity in vitro. So far, this interaction has not been reported in humans. Theoretically, aegeline may increase levels of drugs metabolized by CYP3A4. Some drugs metabolized by CYP3A4 include lovastatin (Mevacor), ketoconazole (Nizoral), itraconazole (Sporanox), fexofenadine (Allegra), triazolam (Halcion), and numerous others. Use aegeline cautiously or avoid in patients taking these drugs.

Here's the honest picture with Anacin (aspirin plus caffeine) and aegeline. The concern on file is that aegeline can slow down a liver enzyme called CYP3A4 in lab dishes. In theory, blocking that enzyme could let certain drugs build up. But this is only in vitro and animal research, and it hasn't been seen in real people. On top of that, aspirin and caffeine aren't mainly cleared by CYP3A4, so this particular pairing is unlikely to cause a meaningful problem.

My bigger worry is aegeline itself. It was pulled from supplements after being tied to serious liver injury, so I'd steer clear of it regardless of your aspirin/caffeine use. Please run any supplement plans by your pharmacist or doctor first.

Aegeline is a compound originally identified in the bael tree (Aegle marmelos) that gained attention because it was added to some weight-loss and workout supplements. It is best known for being linked to serious liver injury in a product called OxyELITE Pro, and U.S. regulators warned against products containing it. There is no good evidence it safely helps with weight loss, and it should generally be avoided.
