Why this applies to Vincristine

Vincristine falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Aegeline interacts with.

Aegeline inhibits cytochrome P450 3A4 (CYP3A4) activity in vitro. So far, this interaction has not been reported in humans. Theoretically, aegeline may increase levels of drugs metabolized by CYP3A4. Some drugs metabolized by CYP3A4 include lovastatin (Mevacor), ketoconazole (Nizoral), itraconazole (Sporanox), fexofenadine (Allegra), triazolam (Halcion), and numerous others. Use aegeline cautiously or avoid in patients taking these drugs.

Here's the short version on combining aegeline with vincristine. Your liver uses an enzyme called CYP3A4 to break down and clear vincristine. In lab dishes, aegeline appears to slow that same enzyme down. If that happened in your body, vincristine could clear more slowly and build up to higher levels, which can mean more of its side effects. With this drug that often means worse nerve problems (tingling, numbness, weakness, constipation) and a bigger drop in blood counts.

That said, this is only test-tube data and has never been confirmed in people, so it's a theoretical worry. The bigger issue is that aegeline itself was linked to serious liver injury and is best avoided altogether. Since vincristine is a powerful chemo drug, please run any supplement past your oncology team or pharmacist first.

Aegeline is a compound originally identified in the bael tree (Aegle marmelos) that gained attention because it was added to some weight-loss and workout supplements. It is best known for being linked to serious liver injury in a product called OxyELITE Pro, and U.S. regulators warned against products containing it. There is no good evidence it safely helps with weight loss, and it should generally be avoided.

What Is Vincristine Injection Used For?

Vincristine Sulfate Injection is approved to treat acute leukemia. It is also used as part of combination chemotherapy regimens for Hodgkin's disease, non-Hodgkin's malignant lymphomas, rhabdomyosarcoma (a soft-tissue cancer), neuroblastoma (a cancer of nerve tissue), and Wilms' tumor (a kidney cancer in children).

Because it works best alongside other cancer medicines, vincristine is almost always given as part of a multi-drug treatment plan rather than on its own.
