Vincristine falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Adrafinil interacts with.

Theoretically, adrafanil might reduce levels and clinical effects of CYP3A4 substrates.
Adrafinil is a precursor to the stimulant prescription drug modafinil. Modafinil has been shown to weakly induce CYP3A4 activity.

Here's the basic idea with these two together. Your liver turns adrafinil into modafinil, and modafinil can gently speed up a liver enzyme called CYP3A4. That same enzyme helps clear vincristine from your body. So in theory, adrafinil could make vincristine break down a little faster, which might lower its levels and weaken how well your chemotherapy works.

I want to be honest about how soft this is. It's a theoretical concern based on how the drugs behave, not something proven in real patients, and modafinil is only a weak inducer. Still, vincristine is serious chemo with a careful dose, so don't combine these on your own. Please run it by your oncologist or pharmacist first.

What Is Vincristine Injection Used For?

Vincristine Sulfate Injection is approved to treat acute leukemia. It is also used as part of combination chemotherapy regimens for Hodgkin's disease, non-Hodgkin's malignant lymphomas, rhabdomyosarcoma (a soft-tissue cancer), neuroblastoma (a cancer of nerve tissue), and Wilms' tumor (a kidney cancer in children).

Because it works best alongside other cancer medicines, vincristine is almost always given as part of a multi-drug treatment plan rather than on its own.
