Aspirin, Carisoprodol, Codeine Phosphate falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES — the group Ayahuasca interacts with.

Theoretically, ayahuasca might increase plasma concentrations of CYP2D6 substrates.
Ayahuasca contains beta-carboline alkaloids, including harmine and harmaline. In vitro research suggests that the beta-carboline alkaloids might inhibit CYP2D6 enzymes. This effect has not been reported in humans.

This combination pill contains codeine, and that's the part worth thinking about here. Codeine is a prodrug, meaning it doesn't do much on its own. Your liver uses an enzyme called CYP2D6 to convert codeine into morphine, which is what actually relieves pain. Lab studies suggest the beta-carboline compounds in ayahuasca (harmine and harmaline) may block CYP2D6.

If that blocking happened in a person, you'd expect less morphine to form, so the codeine might give you weaker pain relief rather than more. To be clear, this is theoretical and only seen in test-tube studies, never confirmed in people. The bigger concern with ayahuasca is its strong MAOI activity, which can dangerously interact with many medicines. Please talk with your pharmacist or doctor before mixing these.
