Daridorexant falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Ayahuasca interacts with.

Theoretically, ayahuasca might increase plasma concentrations of CYP3A4 substrates.
Ayahuasca contains beta-carboline alkaloids. In vitro research suggests that beta-carboline alkaloids competitively inhibit CYP3A4 enzymes. This effect has not been reported in humans.

Daridorexant (Quviviq) is your sleep medicine, and your liver clears it using an enzyme called CYP3A4. The beta-carboline compounds in ayahuasca may slow that same enzyme down, at least based on lab dish studies. If that happened in a real person, your body could clear the daridorexant more slowly, letting it build up. That could mean stronger next-morning grogginess, lingering drowsiness, dizziness, or trouble feeling fully alert.

Here is the honest part: this is theoretical and has only been seen in test-tube research, not in people. Still, ayahuasca carries its own serious risks separate from this. Please talk with your pharmacist or doctor before combining anything with daridorexant.
