Darunavir falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Ayahuasca interacts with.

Theoretically, ayahuasca might increase plasma concentrations of CYP3A4 substrates.
Ayahuasca contains beta-carboline alkaloids. In vitro research suggests that beta-carboline alkaloids competitively inhibit CYP3A4 enzymes. This effect has not been reported in humans.

Here's the gist for darunavir (Prezista): your body relies on an enzyme called CYP3A4 to help process it. The beta-carboline compounds in ayahuasca have shown, in lab dish studies only, that they can block this same enzyme. If that blocking happened in a real person, darunavir could build up to higher levels in your blood, which might mean more side effects like nausea, diarrhea, headache, or rash.

I want to keep this in perspective. This is theoretical, based only on test-tube research, and it has never actually been reported in people. Honestly, the bigger concern with ayahuasca is its MAOI activity, which can clash dangerously with many medicines. Please loop in your doctor or pharmacist before mixing anything with your HIV treatment.

About Darunavir

What Is Darunavir Used For?

Brand name: Prezista

Darunavir treats HIV-1 infection in adults and in children who are at least 3 years old and weigh at least 10 kg (about 22 lbs). It is not a cure for HIV, but it is used as part of a combination antiretroviral regimen to control the virus.

Darunavir must always be taken together with ritonavir — that pairing is required for darunavir to work properly. Both Prezista (brand name) and generic darunavir tablets are approved for this use. Prezista is also available as an oral suspension, which is particularly useful for younger or smaller patients.
