Tamoxifen Citrate falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES — the group Ayahuasca interacts with.

Theoretically, ayahuasca might increase plasma concentrations of CYP2D6 substrates.
Ayahuasca contains beta-carboline alkaloids, including harmine and harmaline. In vitro research suggests that the beta-carboline alkaloids might inhibit CYP2D6 enzymes. This effect has not been reported in humans.

Here's something important about tamoxifen: it's actually a prodrug. Your body has to convert it into its active form (called endoxifen) using an enzyme named CYP2D6. That active form is what does the real work of fighting and preventing breast cancer.

Ayahuasca contains compounds (harmine and harmaline) that may block CYP2D6. If that enzyme is slowed down, your body may make less of the active tamoxifen, which could mean weaker protection against breast cancer. I'll be honest: this concern comes only from lab studies and hasn't been shown in people, so it's theoretical. Still, with a medication this important, I wouldn't take chances. Please talk with your oncologist or pharmacist before mixing the two.

Ayahuasca contains beta-carboline alkaloids. In vitro research suggests that beta-carboline alkaloids competitively inhibit CYP3A4 enzymes. This effect has not been reported in humans.

Here's the wrinkle with tamoxifen: it's actually a prodrug. Your body has to convert it into its active form (mainly endoxifen) using liver enzymes, including CYP3A4, before it can do its job against breast cancer. So this isn't a simple 'levels go up' situation. The beta-carbolines in ayahuasca were shown in lab dish (in vitro) studies to slow CYP3A4, which could in theory shift how tamoxifen is processed and how much active drug you end up with.

I want to be honest about the evidence here: this has only been seen in test-tube research, never in actual people, so it's very theoretical. Still, tamoxifen is a serious cancer medicine you don't want to mess with, and ayahuasca carries its own real risks. Please loop in your oncologist or pharmacist before combining anything.

About Tamoxifen

What Is Tamoxifen Used For?

Brand names: Nolvadex, Soltamox

Tamoxifen is used to treat estrogen receptor-positive metastatic breast cancer in both women and men, and as follow-up (adjuvant) therapy after surgery for early-stage breast cancer in women. It also reduces the occurrence of cancer in the opposite breast when used as adjuvant therapy.

In women with ductal carcinoma in situ (DCIS — an early, non-invasive breast change) following surgery and radiation, tamoxifen lowers the risk of developing invasive breast cancer. It is also approved to reduce the risk of breast cancer in women who are at high risk but have not yet been diagnosed. Both Tamoxifen Citrate tablets and Soltamox oral solution share these indications.
