Dapsone falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Berberine interacts with.

Theoretically, berberine might increase serum levels of drugs metabolized by CYP3A4.
In vitro research and preliminary clinical research show that berberine moderately inhibits CYP3A4. Theoretically, use of berberine with drugs metabolized by CYP3A4 might increase drug levels and increase the risk of adverse effects.

Here's the simple version: your liver uses an enzyme called CYP3A4 to help clear dapsone out of your body. Berberine appears to slow that enzyme down a bit. If it's working less efficiently, dapsone may hang around at slightly higher levels than usual, which could nudge up the chance of side effects.

With dapsone, the side effects worth watching for include unusual tiredness, shortness of breath, a bluish or gray tint to the lips or skin, or yellowing of the eyes, since this drug can stress red blood cells and the liver. This particular concern is still mostly theoretical and based on lab and early research, so don't panic. Just loop in your pharmacist or doctor before adding berberine so they can keep an eye on you.

About Dapsone

What Is Dapsone Used For?

Brand names: Dapsone, Avlosulfon, Aczone

Dapsone tablets are used to treat dermatitis herpetiformis (a chronic, intensely itchy blistering skin condition) and leprosy (all forms, except where proven resistance to dapsone exists). For leprosy, dapsone tablets are typically used in combination with other anti-leprosy medicines.

Aczone Gel (dapsone gel, 7.5%) and dapsone gel are indicated for the topical treatment of acne vulgaris in patients 9 years of age and older. This topical form is applied to the skin and is not used for leprosy or dermatitis herpetiformis.
