Lonapegsomatropin-Tcgd falls under CYTOCHROME P450 3A4 (CYP3A4) INDUCERS — the group Borage interacts with.

Theoretically, taking borage with drugs that induce CYP3A4 might increase levels of pyrrolizidine alkaloid (PA) toxic metabolites.
Although borage seed oil contains little to no PAs, some borage plant parts, such as the leaf, flower, and seed, can contain hepatotoxic PAs. Hepatotoxic PAs are substrates of CYP3A4, which converts these chemicals into toxic metabolites. Tell patients to avoid borage preparations that are not certified and labeled as hepatotoxic PA-free.

Here's the thing to understand: Skytrofa (lonapegsomatropin) is a growth hormone replacement, and it can mildly nudge up the activity of a liver enzyme called CYP3A4. That detail matters because the pyrrolizidine alkaloids (PAs) found in some borage plant parts (leaf, flower, and seed) are turned into their harmful, liver-toxic forms by that very same enzyme. So in theory, speeding up CYP3A4 could mean more of those toxic byproducts.

I want to keep this in perspective though. This concern comes only from lab and animal data, so it's theoretical, not something we've clearly seen in people. The simplest safeguard is choosing borage products clearly labeled PA-free, since proper borage seed oil has little to no PAs. Please run it by your child's doctor or pharmacist first.

Skytrofa (lonapegsomatropin-tcgd) is used to treat growth failure in pediatric patients 1 year and older who weigh at least 11.5 kg (about 25 lbs) and whose bodies do not make enough growth hormone on their own.

Skytrofa is also used to replace growth hormone in adults who have growth hormone deficiency (GHD).
