Ambrisentan falls under HEPATOTOXIC DRUGS — the group Camphor interacts with.

Theoretically, concomitant use of camphor with other hepatotoxic drugs might increase the risk of liver damage.
There have been a few case reports of transient elevations in liver enzymes in adults after oral or topical use of camphor. There has also been one case of hepatotoxicity in an infant after topical camphor use.

Here's the gist: ambrisentan (Letairis) can be hard on the liver, which is why your doctor checks liver enzymes before and during treatment. Camphor, while mostly used as a skin rub, has been linked in a handful of reports to temporary bumps in liver enzymes, even from topical use. The worry is simply that stacking two things that may stress the liver could add up.

I want to be honest about the evidence though: this is theoretical, based on how each works rather than proven cases of the two together. Still, since ambrisentan already carries a liver caution, it's worth playing it safe. Use camphor products only on the skin as directed, never swallow them, and check with your pharmacist or doctor before adding them.

Camphor is a strong-smelling compound from the camphor tree (or made synthetically) that is used in many topical rub-on products for muscle aches, itching, and chest congestion. It can be helpful for these uses when applied to the skin in small, regulated amounts, but it is toxic and potentially deadly if swallowed, so it should never be taken by mouth. Keep camphor products away from children and check with a pharmacist before use.

About Ambrisentan

What Is Ambrisentan Used For?

Brand names: Letairis, Volibris

Ambrisentan tablets and Letairis tablets are both used to treat pulmonary arterial hypertension (PAH, WHO Group 1) — a condition where the blood pressure in the arteries leading to the lungs is dangerously high. Both are approved to help adults exercise better and slow the worsening of PAH.

Both ambrisentan tablets and Letairis are also approved to be used together with tadalafil (another PAH medicine) to further reduce the risk of disease progression, lower the chance of hospitalization for worsening PAH, and improve exercise ability. Most of the clinical evidence was in patients with mild-to-moderate symptoms and PAH caused by unknown or inherited causes, or PAH linked to connective tissue diseases.
