Theoretically, CYP3A4 inhibitors might increase cannabidiol levels.
Cannabidiol is a substrate of CYP3A4 enzymes. Theoretically, drugs that inhibit CYP3A4 enzymes might increase levels of cannabidiol, increasing its effects and adverse effects.

Here's the situation with these two. Your body uses an enzyme called CYP3A4 to help clear CBD out of your system. Amiodarone slows that enzyme down, so in theory it could let CBD build up a bit more than usual. That means the effects and side effects of the CBD (think drowsiness, lightheadedness, diarrhea, or changes in appetite) might feel stronger.

I want to be honest that this is a theoretical concern based on how these drugs behave, not something proven in studies, so don't panic. Still, amiodarone is a serious heart-rhythm drug with a narrow safety margin and its own long list of interactions, so I'd really want you to check with your pharmacist or cardiologist before adding any CBD product.

In vitro and animal research shows that cannabidiol inhibits CYP3A4. In human studies and case reports, cannabidiol has been associated with an increase in plasma levels of the CYP3A4 substrates zonisamide, tacrolimus, everolimus, citalopram, midazolam, and methadone.

Here's the gist: your body uses a liver enzyme called CYP3A4 to help clear amiodarone. CBD can slow that enzyme down, so amiodarone may build up more than usual in your system. Amiodarone is already an active drug (not a prodrug that needs switching on), so higher levels generally mean stronger effects and a bigger chance of side effects.

With amiodarone that's worth respecting, because it has a narrow safety window and a very long stay in the body. Watch for things like a slow or irregular heartbeat, dizziness, fatigue, or feeling faint. Please don't change anything on your own. Loop in your pharmacist or doctor before combining these, since they may want to monitor you more closely.

Theoretically, cannabidiol might increase levels of drugs metabolized by CYP2C8.
In vitro research shows that cannabidiol inhibits CYP2C8. However, this interaction has yet to be reported in humans. Until more is known, use with caution. Theoretically, concomitant use of cannabidiol with CYP2C8 substrates might increase the risk for adverse effects from these substrates.

Here's the gist for you. Amiodarone is one of the medicines your liver clears partly through an enzyme called CYP2C8. In lab (test tube) studies, CBD appears to slow that same enzyme down. If that happens in the body too, amiodarone could build up a bit more than expected, which might mean stronger effects and a higher chance of side effects like an overly slow heartbeat, dizziness, tiredness, or stomach upset.

That said, this is theoretical. It has only been seen in lab research, not in real people, so we don't actually know if it matters with your dose. But amiodarone is a tricky, narrow-margin heart drug where small shifts can count, so please don't add CBD on your own. Run it by your pharmacist or doctor first.

Amiodarone is prescribed for adults with serious, life-threatening heart-rhythm problems that haven't responded to other medications. Specifically, it's used for two dangerous arrhythmias (abnormal heartbeats): recurrent ventricular fibrillation (a chaotic, potentially fatal heart rhythm) and recurrent ventricular tachycardia (a dangerously fast heart rhythm that causes unstable blood pressure).
