Siltuximab falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Dhea interacts with.

Theoretically, DHEA might increase the levels of drugs metabolized by CYP3A4.
Some preliminary evidence shows that DHEA may inhibit CYP3A4; however, the clinical significance of this potential interaction is not known.

Here's the honest take: this interaction is mostly a theoretical concern on paper, and it doesn't fit Siltuximab very well. DHEA may slightly block an enzyme called CYP3A4, which can cause some medications to build up. But Siltuximab (Sylvant) is a monoclonal antibody, a large protein given by IV. Antibodies like this aren't broken down by CYP3A4 the way ordinary pills are, so I wouldn't expect DHEA to directly raise its levels or its side effects.

The bigger picture is that Siltuximab can actually change how your liver handles other CYP3A4 medications after your inflammation calms down. Since DHEA is a hormone with real effects of its own, please run it past your oncology team or pharmacist before combining the two.

What Is Siltuximab Injection Used For?

Brand name: Sylvant

Sylvant (siltuximab) injection is approved to treat multicentric Castleman's disease (MCD) — a rare disorder involving abnormal lymph node growth driven by an overactive immune signal called IL-6. It is specifically approved for patients who test negative for both HIV and HHV-8 (human herpesvirus-8).

Sylvant has not been studied in patients with MCD who are HIV-positive or HHV-8-positive, because it does not block the form of IL-6 produced by those viruses.
