Vemurafenib falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Passion Flower interacts with.

Theoretically, passion flower might decrease the effects of CYP3A4 substrates.
In vitro research suggests that passion flower can induce CYP3A4 enzymes, albeit to a much lower degree than rifampin, a known CYP3A4 inducer.

Here's the gist for vemurafenib (Zelboraf). Your body relies partly on a liver enzyme called CYP3A4 to process this cancer medicine. Lab-dish studies hint that passion flower may speed that enzyme up a little, which in theory could clear vemurafenib out of your system faster and make it slightly less effective at fighting the melanoma or Erdheim-Chester disease.

I want to be straight with you: this concern comes only from test-tube research, not from people, and the effect was far weaker than with strong inducers. So it's more of a theoretical caution than a proven problem. Still, because vemurafenib is doing serious work, I'd run any passion flower use past your oncologist or pharmacist before starting it.

About Vemurafenib

What Is Vemurafenib Used For?

Brand name: Zelboraf

Zelboraf (vemurafenib) is used to treat adults with unresectable or metastatic melanoma — meaning melanoma that cannot be surgically removed or has spread — when the tumor tests positive for a specific genetic change called the BRAF V600E mutation. An FDA-approved test must confirm this mutation before starting treatment.

Zelboraf is also approved for patients with Erdheim-Chester Disease (ECD), a rare condition where abnormal immune cells build up in organs, when that disease carries a BRAF V600 mutation. Zelboraf is not for patients whose melanoma has normal (wild-type) BRAF, as it may actually promote tumor growth in that setting.
