Lorlatinib falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Rooibos interacts with.

Theoretically, taking rooibos with CYP3A4 substrates may increase the effects of CYP3A4 substrates.
In vitro research shows that the methanol extract of rooibos leaves and stems strongly inhibits CYP3A4 enzyme activity.

Here's the gist with rooibos and Lorbrena (lorlatinib). Your body uses an enzyme called CYP3A4 to clear lorlatinib out of your system. In lab studies, a concentrated rooibos extract slowed that enzyme down. If that happened in a person, lorlatinib could clear out more slowly and build up higher than expected, which may mean more of its side effects, things like high cholesterol or triglycerides, swelling, mood or memory changes, fatigue, or nerve tingling.

The good news: this is only a test-tube finding, not something proven in people, and a normal cup of rooibos tea is very different from a strong extract. Since lorlatinib is a serious cancer drug, please don't start or stop anything on your own. Just run rooibos tea or extracts past your oncologist or pharmacist first.

What Is Lorlatinib Used For?

Brand name: Lorbrena

Lorbrena (lorlatinib) is used to treat adults with metastatic non-small cell lung cancer (NSCLC) — meaning lung cancer that has spread — when lab testing confirms the tumor is ALK-positive (has a specific gene change that drives cancer growth). Patients must be tested with an FDA-approved diagnostic test before starting treatment.

Lorbrena has been studied in patients who have not yet received treatment for advanced disease, as well as in patients whose cancer progressed on other ALK-targeting medicines.
