These two interact in 2 distinct ways. Each one is explained separately below.
Why Are There 2 Interactions?
Sulforaphane interacts with whole categories of medication. Aspirin, Caffeine, Orphenadrine falls into 2 of those categories, so it can interact with Sulforaphane in 2 different ways. We’ve listed each one below — your overall risk is driven by the most serious of them (Moderate).
CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES
1 of 2Why this applies to Aspirin, Caffeine, Orphenadrine
Aspirin, Caffeine, Orphenadrine falls under CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES — the group Sulforaphane interacts with.
Animal and in vitro research shows that sulforaphane inhibits CYP1A2 enzyme activity. However, its precursor glucoraphanin also appears to increase the expression of the CYP1A2 enzyme, which could lead to increased metabolism of CYP1A2 substrates. These effects have not been reported in humans.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
This combination product blends aspirin, caffeine, and a muscle relaxer (orphenadrine). The one piece that overlaps with sulforaphane is the caffeine, which your body clears mainly through an enzyme called CYP1A2. In lab and animal studies, sulforaphane can nudge this enzyme in two opposite directions, so it's genuinely hard to predict which way things go in a real person.
If the enzyme is slowed, caffeine could linger longer, meaning a bit more jitteriness, a faster heartbeat, or trouble sleeping. The good news: this is theoretical only and hasn't been shown in humans, and broccoli on your plate won't matter. With a concentrated sulforaphane supplement, it's worth a quick chat with your pharmacist to be safe.

CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES
2 of 2Aspirin, Caffeine, Orphenadrine also falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES.
Animal and in vitro research shows that sulforaphane inhibits CYP3A4 enzyme activity and downregulates its expression. This interaction has not been reported in humans.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
This combination pill contains three active ingredients, and the one that matters most here is orphenadrine, the muscle relaxant. Your liver uses an enzyme called CYP3A4 to help break it down. In lab and animal studies, sulforaphane appears to slow that enzyme. In theory, if orphenadrine clears more slowly, its levels could creep up, which might make side effects like dry mouth, blurry vision, dizziness, drowsiness, or a fast heartbeat more noticeable.
Here is the important part: this is only seen in test tubes and animals, not in people, so the real-world risk looks low. Broccoli at dinner is no concern. A concentrated sulforaphane supplement is a different story, so check with your pharmacist or doctor before starting one.

About Sulforaphane
What is Sulforaphane?
- Antioxidant support
- General cellular health
- Detoxification support
- Inflammation
- Skin and aging concerns
Sulforaphane is a natural compound found in broccoli and other cruciferous vegetables that has shown promising antioxidant and anti-inflammatory effects in lab studies, but strong human evidence for most specific health claims is still limited. It is generally well tolerated as part of a vegetable-rich diet, but if you take a concentrated supplement, talk to your pharmacist or doctor first.
