Belladonna, Caffeine, Ergotamine, Pentobarbital falls under CNS DEPRESSANTS — the group Tree Turmeric interacts with.
Animal research suggests that berberine, a constituent of tree turmeric, can have sedative effects.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
This combination pill includes pentobarbital, a barbiturate that calms the nervous system, which is the part that matters here. In animal studies, berberine (the active compound in tree turmeric) showed mild sedative effects of its own. So the concern is simple addition: taking the two together might make you feel more drowsy, more lightheaded, or slower to react than the medicine alone. You could notice extra grogginess or trouble concentrating.
Keep in mind this is theoretical, based only on lab and animal research, so I wouldn't lose sleep over it. Still, since pentobarbital already causes sedation, it's worth a quick chat with your pharmacist or doctor before adding a berberine supplement, especially before driving.

CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES

In vitro research and preliminary clinical evidence suggest that berberine, a constituent of tree turmeric, moderately inhibits CYP3A4.
Interaction has not been documented in well-controlled studies, however, the interaction has been demonstrated in some small human studies or in controlled animal studies in conjunction with multiple case reports..
This combination pill includes ergotamine, which your liver clears largely through an enzyme called CYP3A4. Berberine, the active part of tree turmeric, can moderately slow that same enzyme. If ergotamine breaks down more slowly, it can build up, and that matters because ergot drugs are sensitive. Too much can cause cold, numb, or tingling hands and feet, leg cramps, nausea, or a pounding headache.
The caffeine and pentobarbital pieces can also feel stronger, meaning more jitteriness or more drowsiness. The evidence here is mostly lab-based and theoretical, so it is not a guarantee, but ergotamine deserves caution. Please don't start or stop anything on your own. Just check with your pharmacist or doctor first.

PENTOBARBITAL (Nembutal)

Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
This combination product contains pentobarbital, a barbiturate that calms the brain and can make you drowsy. The concern here is with that ingredient. In animal studies, berberine (the active compound in tree turmeric) made pentobarbital-induced sleep last longer. So the worry is that tree turmeric could deepen or stretch out the sedative effect of the pentobarbital part of this medicine.
If that played out in people, you might feel extra sleepy, groggy, dizzy, or slow to wake up. I'll be honest with you though: this is based on lab and animal work, not human studies, so it's more of a theoretical caution than a proven problem. Even so, since pentobarbital is potent, it's worth running this past your pharmacist or doctor before combining them.
