Acetaminophen, Butalbital, Codeine Phosphate falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES — the group Ayahuasca interacts with.

Theoretically, ayahuasca might increase plasma concentrations of CYP2D6 substrates.
Ayahuasca contains beta-carboline alkaloids, including harmine and harmaline. In vitro research suggests that the beta-carboline alkaloids might inhibit CYP2D6 enzymes. This effect has not been reported in humans.

Here's the wrinkle with this combo product: the codeine in it is a prodrug. Your body has to use an enzyme called CYP2D6 to turn codeine into morphine, which is what actually relieves pain. Lab studies suggest the beta-carboline compounds in ayahuasca (harmine and harmaline) may block CYP2D6. If that happens, less codeine gets converted to morphine, so you might get weaker pain relief from this medication.

I want to be honest: this is only seen in test-tube research, not in people, so it's theoretical. The bigger worry is ayahuasca itself, since its MAOI activity can cause dangerous reactions with many drugs. Please talk with your pharmacist or doctor before mixing these.
