Why this applies to Dextroamphetamine Sulphate

Dextroamphetamine Sulphate falls under SEROTONERGIC DRUGS — the group Ayahuasca interacts with.

Theoretically, ayahuasca might increase levels of serotonin, which may have additive effects with serotonergic drugs.
In vitro and animal research shows that harmaline and harmine, constituents of ayahuasca, strongly and reversibly inhibit monoamine oxidase A and weakly inhibit monoamine oxidase B. The dimethyltryptamine and tetrahydroharmine constituents of ayahuasca have also demonstrated serotonergic activity. A case of serotonin syndrome characterized by tremors, sweating, shivering, confusion, nausea, vomiting, and disorientation has been reported in a 36-year-old male who consumed 100 mL of ayahuasca while taking fluoxetine 20 mg daily. Combining serotonergic drugs with ayahuasca might increase the risk of serotonergic side effects including serotonin syndrome and cerebral vasoconstrictive disorders.

Here's the concern with mixing ayahuasca and your dextroamphetamine (Dexedrine). The vine in ayahuasca acts as a strong MAO inhibitor, which means it blocks the enzyme your body uses to break down certain brain chemicals. Dextroamphetamine works by flooding your system with those same chemicals (dopamine, norepinephrine, and some serotonin). When you can't clear them normally, they can pile up to dangerous levels.

This combination can push blood pressure dangerously high and, on the serotonin side, trigger serotonin syndrome (agitation, sweating, shivering, fast heartbeat, confusion, tremors). The MAOI-plus-stimulant combo is one pharmacists specifically warn against. Even though this is rated theoretical, the risk is serious enough that I would not combine them. Please talk with your doctor or pharmacist first.

Dextroamphetamine Sulphate also falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES.

Theoretically, ayahuasca might increase plasma concentrations of CYP2D6 substrates.
Ayahuasca contains beta-carboline alkaloids, including harmine and harmaline. In vitro research suggests that the beta-carboline alkaloids might inhibit CYP2D6 enzymes. This effect has not been reported in humans.

Here's the gist for dextroamphetamine: ayahuasca's beta-carboline compounds (harmine and harmaline) may slow down the CYP2D6 enzyme, which is one of the pathways your body uses to clear this stimulant. If that pathway gets blocked, more of the drug could linger in your system, possibly amplifying its effects like a racing or pounding heart, higher blood pressure, jitteriness, trouble sleeping, and reduced appetite.

Honestly though, my bigger worry is a different one. The MAOI part of ayahuasca combined with a stimulant like dextroamphetamine can push norepinephrine and serotonin dangerously high, which can lead to a serious blood pressure spike or serotonin reaction. The CYP2D6 piece here is only from lab studies, not people, but please don't pair these without talking to your doctor or pharmacist first.
