Dorzolamide, Timolol falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES — the group Ayahuasca interacts with.

Theoretically, ayahuasca might increase plasma concentrations of CYP2D6 substrates.
Ayahuasca contains beta-carboline alkaloids, including harmine and harmaline. In vitro research suggests that the beta-carboline alkaloids might inhibit CYP2D6 enzymes. This effect has not been reported in humans.

Let me put this one in plain terms. Your Cosopt eye drops contain timolol, which is a beta blocker that the liver clears using an enzyme called CYP2D6. The beta-carboline compounds in ayahuasca (harmine and harmaline) may slow that same enzyme down. If timolol clears more slowly, a bit more of it can build up in your body, which could nudge your heart rate lower or drop your blood pressure, and may cause tiredness or lightheadedness.

That said, this is based only on lab studies, not on real people, so it stays theoretical. Even though eye drops put only a small amount into your bloodstream, ayahuasca is potent and risky on its own. Please loop in your doctor or pharmacist before mixing the two.

What Is Dorzolamide and Timolol Ophthalmic Used For?

Brand name: Cosopt

Dorzolamide and timolol ophthalmic eye drops are used to reduce elevated intraocular pressure (pressure inside the eye) in people with open-angle glaucoma or ocular hypertension (higher-than-normal eye pressure without signs of glaucoma damage). It is specifically intended for patients who have not responded well enough to beta-blocker eye drops alone.

This combination is available as Cosopt, Cosopt PF, and Dorzolamide Hydrochloride and Timolol Maleate eye drops. Cosopt PF is the preservative-free formulation, which may be preferred for people sensitive to preservatives.
