Propoxyphene Napsylate falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES — the group Ayahuasca interacts with.

Theoretically, ayahuasca might increase plasma concentrations of CYP2D6 substrates.
Ayahuasca contains beta-carboline alkaloids, including harmine and harmaline. In vitro research suggests that the beta-carboline alkaloids might inhibit CYP2D6 enzymes. This effect has not been reported in humans.

Here's the gist for Darvon N (propoxyphene napsylate). Your body relies partly on an enzyme called CYP2D6 to break this opioid down. The beta-carboline compounds in ayahuasca (harmine and harmaline) can block that enzyme, at least in lab studies. If that happens in a real person, propoxyphene could clear more slowly and build up, which may strengthen its effects and raise the odds of side effects like drowsiness, dizziness, confusion, nausea, slowed breathing, or heart rhythm trouble.

I want to be honest though: this is theoretical, based only on test-tube work, and hasn't been confirmed in people. On top of that, ayahuasca's MAOI activity makes mixing it with opioids genuinely risky on its own. Please don't combine these without talking to your doctor or pharmacist first.
