Ramelteon falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Ayahuasca interacts with.

Theoretically, ayahuasca might increase plasma concentrations of CYP3A4 substrates.
Ayahuasca contains beta-carboline alkaloids. In vitro research suggests that beta-carboline alkaloids competitively inhibit CYP3A4 enzymes. This effect has not been reported in humans.

Here's the basic idea with Rozerem (ramelteon) and ayahuasca. Your liver uses an enzyme called CYP3A4 to clear ramelteon out of your body. The beta-carboline compounds in ayahuasca (like harmine) appear to slow that same enzyme down, at least in lab dish testing. If that happens in a real person, ramelteon could build up to higher levels than usual, which might mean a stronger sedating effect, more next-morning grogginess, dizziness, or feeling unsteady on your feet.

That said, I want to be honest about the evidence. This is theoretical and based only on lab studies, not on people, so we don't truly know how big a deal it is. Ayahuasca also carries its own serious risks on top of this. Please check with your doctor or pharmacist before mixing the two.
