Loperamide, Simethicone falls under ANTICHOLINERGIC DRUGS — the group Datura Wrightii interacts with.

Datura wrightii contains atropine and scopolamine, as well as other tropane alkaloids with anticholinergic activity. Concomitant use of Datura wrightii with anticholinergic drugs might increase anticholinergic effects and adverse effects. Some anticholinergic drugs include amantadine, atropine, belladonna alkaloids, phenothiazines, scopolamine, and tricyclic antidepressants.

First things first: I really don't want you taking Datura wrightii at all. It's genuinely poisonous, and even small amounts can cause severe harm. So please don't think of this as a normal supplement pairing.

Here's the concern with Imodium Advanced (loperamide plus simethicone). Datura is loaded with atropine and scopolamine, which have strong anticholinergic effects (they dry you out and slow your gut). Loperamide already works by slowing the bowel down, so stacking the two could pile on side effects like a dangerously dry mouth, blurry vision, a racing heart, trouble urinating, and confusion. This is a theoretical concern based on how these compounds work, not hard clinical proof, but the bigger danger is Datura itself. Please talk with your pharmacist or doctor before combining anything like this.
