Here's the gist: DHEA is a hormone supplement, and adding it on top of an antidepressant like amitriptyline may, in theory, nudge your mood in an unexpected direction. The worry isn't about DHEA changing how your body breaks the drug down. Instead, it's that both can affect brain chemistry, and the combo might raise the odds of mood-related side effects such as feeling unusually wired, agitated, restless, or in rare cases a manic-type swing (racing thoughts, very high energy, trouble sleeping).

I want to be honest about the evidence: this is mostly theoretical, with one case report involving a different type of antidepressant. So it's a reasonable caution, not a red alert. If you're considering DHEA while on amitriptyline, please run it by your pharmacist or doctor first, especially since DHEA acts as a real hormone.

Some preliminary evidence shows that DHEA may inhibit CYP3A4; however, the clinical significance of this potential interaction is not known.

Here's the gist for amitriptyline (Elavil). Your liver uses an enzyme called CYP3A4 to help clear this drug out of your system. There's some early, fairly weak evidence that DHEA may slow that enzyme down a bit. If it does, amitriptyline could linger a little longer than usual, which in theory might nudge up its typical side effects, things like drowsiness, dry mouth, constipation, blurry vision, dizziness when standing, or a faster heartbeat.

I want to be straight with you though: this is mostly theoretical and based on scattered reports, not solid studies, so the real-world impact is unknown and may be small. Still, since amitriptyline can be sensitive, it's worth a quick chat with your pharmacist or doctor before pairing the two, rather than guessing on your own.
