Lorlatinib falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Dhea interacts with.

Theoretically, DHEA might increase the levels of drugs metabolized by CYP3A4.
Some preliminary evidence shows that DHEA may inhibit CYP3A4; however, the clinical significance of this potential interaction is not known.

Here's the basic idea with these two together. Your body uses an enzyme called CYP3A4 to break down lorlatinib. Some early lab hints suggest DHEA might slow that same enzyme down. If that happened, lorlatinib could clear out of your body a bit more slowly and build up, which in theory could mean more of its known side effects, like high cholesterol or triglycerides, swelling, mood or thinking changes, fatigue, or numbness and tingling.

I want to be honest though: this is very weak, theoretical evidence, and we don't really know if it matters in real people. Since lorlatinib is a serious cancer medicine, please don't start or stop anything on your own. Just run the idea of adding DHEA past your oncologist or pharmacist first.

What Is Lorlatinib Used For?

Brand name: Lorbrena

Lorbrena (lorlatinib) is used to treat adults with metastatic non-small cell lung cancer (NSCLC) — meaning lung cancer that has spread — when lab testing confirms the tumor is ALK-positive (has a specific gene change that drives cancer growth). Patients must be tested with an FDA-approved diagnostic test before starting treatment.

Lorbrena has been studied in patients who have not yet received treatment for advanced disease, as well as in patients whose cancer progressed on other ALK-targeting medicines.
