CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES
Why this applies to Zopiclone
Zopiclone falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Gymnema interacts with.
One in vitro study using rat liver microsomes shows that gymnema can modestly inhibit the CYP3A4 enzyme. However, other in vitro research using human liver microsomes shows that gymnema does not affect CYP3A4 activity. Animal research also shows that gymnema does not alter the function of CYP3A4. In one study in rats, oral administration of gymnema for 7 days did not alter the clearance of amlodipine, a CYP3A4 substrate.
Interaction has been demonstrated in animal or in lab research but has been shown not to occur in humans..
Here's the short version: your body uses an enzyme called CYP3A4 to help clear zopiclone out of your system. The thinking is that gymnema might slow that enzyme down a bit, which could let zopiclone build up. If that happened, you might notice stronger sleepiness, next-morning grogginess, dizziness, or that metallic, bitter taste zopiclone is known for feeling more pronounced.
That said, I wouldn't lose sleep over this one. The only hint of a problem comes from a test-tube study using rat liver, while human-tissue and live animal studies found no real effect on this enzyme. So it's mostly theoretical. Still, since both can make you drowsy, it's worth a quick chat with your pharmacist before combining them.
