Cyclobenzaprine falls under CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES — the group Gymnema interacts with.

Theoretically, gymnema might increase levels of drugs metabolized by CYP1A2.
Animal and in vitro research shows that gymnema can inhibit the CYP1A2 enzyme. In one animal study, oral administration of gymnema for 7 days increased the plasma concentrations of phenacetin, a CYP1A2 substrate, by about 1.4-fold and reduced the clearance of phenacetin by about 29%.

Here's the basic idea with Gymnema and cyclobenzaprine. Your body relies on a liver enzyme called CYP1A2 to break down cyclobenzaprine and clear it out. Gymnema appears to slow that enzyme down, at least in lab and animal studies. If it does the same in people, cyclobenzaprine could linger a bit longer and build up, which may nudge up its usual side effects like drowsiness, dizziness, dry mouth, and grogginess.

I want to be honest about the strength of this, though. It comes only from test tube and animal research, not from real people taking these two together, so it stays theoretical. Still, since cyclobenzaprine already makes many folks sleepy, it's worth a quick chat with your pharmacist or doctor before combining them, especially if you take other sedating medicines.

Theoretically, gymnema might increase levels of drugs metabolized by CYP3A4.
One in vitro study using rat liver microsomes shows that gymnema can modestly inhibit the CYP3A4 enzyme. However, other in vitro research using human liver microsomes shows that gymnema does not affect CYP3A4 activity. Animal research also shows that gymnema does not alter the function of CYP3A4. In one study in rats, oral administration of gymnema for 7 days did not alter the clearance of amlodipine, a CYP3A4 substrate.

Here's the short version: your body clears cyclobenzaprine in part through a liver enzyme called CYP3A4. The thought is that gymnema might slow that enzyme down a touch, which in theory could let a little more cyclobenzaprine linger in your system. If that happened, you could feel its usual effects more strongly, things like drowsiness, dry mouth, dizziness, or feeling foggy.

That said, I wouldn't lose sleep over this one. The concern comes from a single test-tube study using rat liver, while studies in human liver tissue and live animals showed no real effect. So this is a very weak, mostly theoretical interaction. Still, since cyclobenzaprine already makes many people sleepy, it's worth a quick chat with your pharmacist before pairing the two.

What Is Cyclobenzaprine Used For?

Brand names: Cyclobenzaprine, Flexeril, Cycloprine

Cyclobenzaprine tablets and extended-release capsules are used alongside rest and physical therapy to relieve muscle spasm caused by acute, painful musculoskeletal conditions — easing pain, tenderness, and limited movement. This use is intended for short-term treatment only (up to 2–3 weeks).

Tonmya, the sublingual (under-the-tongue) form of cyclobenzaprine, is separately indicated for the treatment of fibromyalgia in adults. It is not the same as the oral tablet or capsule formulations.
