Agomelatine falls under CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES — the group Ipriflavone interacts with.

Ipriflavone is thought to competitively inhibit cytochrome P450 1A2 (CYP1A2). Theoretically, concurrent use of ipriflavone and drugs metabolized by these enzymes might result in decreased drug elimination, increased serum levels, and potential toxicity. Some drugs metabolized by CYP1A2 include clozapine (Clozaril), cyclobenzaprine (Flexeril), fluvoxamine (Luvox), haloperidol (Haldol), imipramine (Tofranil), mexiletine (Mexitil), olanzapine (Zyprexa), Pentazocine (Talwin), propranolol (Inderal), tacrine (Cognex), theophylline (Slo-bid, Theo-Dur, others), zileuton (Zyflo), Zolmitriptan (Zomig), and others.

Here's the gist for Agomelatine (Valdoxan): your body relies heavily on a liver enzyme called CYP1A2 to clear this antidepressant out of your system. Ipriflavone is thought to slow that same enzyme down. So in theory, pairing them could let agomelatine build up higher than expected, which might mean more dizziness, drowsiness, headache, nausea, or a bit more strain on the liver.

I'll be honest with you though, this concern comes from lab studies only, not real people, so we don't truly know how big a deal it is. Agomelatine isn't a prodrug, so the math here is simply more drug hanging around. Since agomelatine already needs liver monitoring, it's worth a quick chat with your pharmacist or doctor before combining the two.

Here's the gist for Agomelatine (Valdoxan). Your liver uses an enzyme called CYP2C9 to help clear this antidepressant out of your system. Ipriflavone may gently block that enzyme, which in theory could let agomelatine build up a bit higher than usual.
