Chlorpheniramine, Phenylephrine, Pyrilamine falls under SEROTONERGIC DRUGS — the group Psilocybin interacts with.

Theoretically, combining psilocybin with serotonergic drugs might cause additive serotonergic effects.
Psilocybin has serotonin 2A (5-HT2A) receptor agonist activity. Theoretically, combining serotonergic drugs with psilocybin might increase the risk of serotonergic side effects, including serotonin syndrome. However, this interaction has not been reported in humans.

This combo cold-and-allergy product (Tanamine) contains chlorpheniramine, which gently nudges up serotonin activity in addition to drying you out and making you sleepy. Psilocybin works strongly on serotonin (the 5-HT2A receptor), so the concern on file is simply that stacking two serotonin-acting substances could add up and raise the small risk of too much serotonin, what we call serotonin syndrome (agitation, racing heart, sweating, shivering, or confusion).

To be honest, this is a theoretical worry based on how the drugs work, not something reported in real patients, and the serotonin effect from chlorpheniramine is mild. Still, since psilocybin is investigational and best left to a supervised setting, please run any plans past your pharmacist or doctor first.

In this combination product, the ingredient that matters most here is phenylephrine, a decongestant that gently revs up the body by tightening blood vessels. Psilocybin also has stimulating effects on the nervous system. The concern on file is that stacking these two together could add up and make stimulant-type side effects more likely, things like a faster or pounding heartbeat, higher blood pressure, jitteriness, headache, or trouble sleeping.

I want to be honest about how strong this evidence is: it is theoretical, based on how these substances work rather than real reported cases. So it is not a reason to panic. That said, psilocybin is investigational and illegal in most places, and should only ever be used in a supervised setting. If this is something you are considering, please talk it through with your doctor or pharmacist first.
