Tapentadol falls under SEROTONERGIC DRUGS — the group Psilocybin interacts with.

Theoretically, combining psilocybin with serotonergic drugs might cause additive serotonergic effects.
Psilocybin has serotonin 2A (5-HT2A) receptor agonist activity. Theoretically, combining serotonergic drugs with psilocybin might increase the risk of serotonergic side effects, including serotonin syndrome. However, this interaction has not been reported in humans.

Here's the thing about tapentadol: besides easing pain, it raises serotonin levels in your nervous system. Psilocybin (really its active form, psilocin) also stirs up serotonin activity by acting on certain brain receptors. Stack the two together and you've got two things pushing on the same serotonin system, which in theory could add up.

I want to be honest about how solid this is. This concern comes from how these drugs work on paper, not from real reports in people. Still, if too much serotonin builds up, you can get what's called serotonin syndrome: agitation, fast heartbeat, sweating, shivering, muscle twitching, or confusion. That can be serious. Psilocybin also isn't an approved supplement and is illegal in most places. Please talk with your doctor or pharmacist before mixing anything with tapentadol.

About Tapentadol

What Is Tapentadol Used For?

Brand names: Nucynta, Nucynta ER

Tapentadol tablets (including Nucynta and Tapentadol Hydrochloride tablets) are used for severe acute pain in adults when other treatments are not adequate. Nucynta tablets are also approved for acute pain in children aged 6 and older who weigh at least 40 kg.

Nucynta ER and tapentadol extended-release tablets are used for severe, ongoing pain that requires around-the-clock opioid treatment and cannot be managed with other options, including severe neuropathic pain (nerve pain) caused by diabetic peripheral neuropathy in adults. Extended-release formulations are not intended for as-needed use.
