These two interact in 5 distinct ways. Each one is explained separately below.
Why Are There 5 Interactions?
Rooibos interacts with whole categories of medication. Amitriptyline, Chlordiazepoxide falls into 5 of those categories, so it can interact with Rooibos in 5 different ways. We’ve listed each one below — your overall risk is driven by the most serious of them (Moderate).
CYTOCHROME P450 2C19 (CYP2C19) SUBSTRATES
1 of 5Why this applies to Amitriptyline, Chlordiazepoxide
Amitriptyline, Chlordiazepoxide falls under CYTOCHROME P450 2C19 (CYP2C19) SUBSTRATES — the group Rooibos interacts with.
In vitro research shows that the methanol extract of rooibos leaves and stems strongly inhibits CYP2C19 enzyme activity.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Limbitrol DS combines amitriptyline (an antidepressant) with chlordiazepoxide (a calming benzodiazepine). Both rely partly on liver enzymes, including CYP2C19, to be broken down and cleared. In test-tube studies, rooibos appears to slow that enzyme. If it does the same in people, these two medicines could hang around longer and feel a bit stronger than expected. That might mean more drowsiness, dizziness, dry mouth, constipation, blurred vision, or feeling foggy and unsteady.
Here's the reassuring part: this is only laboratory evidence, not something proven in humans, and a regular cup of rooibos tea is unlikely to cause trouble. Concentrated rooibos extracts are the bigger question mark. If you're taking Limbitrol DS, just check with your pharmacist or doctor before using rooibos supplements.

CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES
2 of 5Amitriptyline, Chlordiazepoxide also falls under CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES.
In vitro research shows that the methanol extract of rooibos leaves and stems inhibits CYP1A2 enzyme activity.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the gist with Limbitrol DS (the amitriptyline plus chlordiazepoxide combo). Lab studies hint that rooibos may slow down a liver enzyme called CYP1A2, which helps clear amitriptyline. If that enzyme is blocked, a little more of the drug could hang around than usual. In plain terms, you might feel more of its common effects, like drowsiness, dry mouth, constipation, blurry vision, or feeling lightheaded when you stand up.
That said, this is only a theoretical concern from test-tube research, not from people, and a normal cup or two of rooibos tea is unlikely to cause trouble. Concentrated rooibos extracts are a bigger question mark. Before adding those, just check with your pharmacist or doctor.
- Fantoukh OI, Dale OR, Parveen A, et al. Safety Assessment of Phytochemicals Derived from the Globalized South African Rooibos Tea (Aspalathus linearis) through Interaction with CYP, PXR, and P-gp. J Agric Food Chem. 2019;67(17):4967-4975.
CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES
3 of 5Amitriptyline, Chlordiazepoxide also falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES.
In vitro research shows that the methanol extract of rooibos leaves and stems inhibits CYP2D6 enzyme activity.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the gist for Limbitrol DS, which combines amitriptyline (an older antidepressant) with chlordiazepoxide (a sedative). The amitriptyline part is partly broken down by a liver enzyme called CYP2D6. In test-tube studies, rooibos appears to slow that enzyme down a bit. In theory, that could let amitriptyline build up slightly, which might mean more of its usual effects like drowsiness, dry mouth, constipation, blurry vision, or a faster heartbeat.
Now, the honest part: this is based only on lab research, not real people, so the actual risk from drinking rooibos as a tea is likely very small. I wouldn't lose sleep over a cup or two. If you're using concentrated rooibos extracts, or you notice extra grogginess, just check in with your pharmacist or doctor.
CYTOCHROME P450 2C9 (CYP2C9) SUBSTRATES
4 of 5Amitriptyline, Chlordiazepoxide also falls under CYTOCHROME P450 2C9 (CYP2C9) SUBSTRATES.
In vitro research shows that the methanol extract of rooibos leaves and stems inhibits CYP2C9 enzyme activity.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Limbitrol DS combines amitriptyline (an antidepressant) with chlordiazepoxide (a sedative in the same family as Valium). The thinking here is that compounds in rooibos may slow down a liver enzyme called CYP2C9, which helps clear these medicines. If that enzyme works more slowly, a little more of the drug could linger in your body, which might mean stronger effects like extra drowsiness, dizziness, dry mouth, constipation, or feeling groggy.
That said, this is a very soft signal. It comes only from lab studies on rooibos extract, not from real people, and a cup or two of rooibos tea is unlikely to cause trouble for most folks. Concentrated rooibos extract supplements are the bigger question mark. Before adding those, just run it by your pharmacist or doctor.

CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES
5 of 5Amitriptyline, Chlordiazepoxide also falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES.
In vitro research shows that the methanol extract of rooibos leaves and stems strongly inhibits CYP3A4 enzyme activity.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Limbitrol DS combines two active medicines: amitriptyline (an antidepressant) and chlordiazepoxide (a benzodiazepine for anxiety). Both are partly broken down by a liver enzyme called CYP3A4. In test-tube studies, a concentrated rooibos extract seemed to slow that enzyme down. If that happened in your body, these medicines could clear more slowly and build up, which might mean more drowsiness, dizziness, dry mouth, constipation, blurred vision, or feeling foggy or unsteady.
Here's the reassuring part: this is only lab-dish evidence, not proof from people, and a regular cup of rooibos tea is very different from a strong supplement extract. Enjoy the tea if you like it, but check with your pharmacist or doctor before using concentrated rooibos products.

From our pharmacist’s overview on the Amitriptyline, Chlordiazepoxide page
“Alcohol is the first thing to know about here. Beer and wine both carry major, well-documented interactions with Amitriptyline and Chlordiazepoxide (Limbitrol DS), because both drugs already slow the central nervous system, and anything sedating layered on top can deepen ordinary drowsiness into something dangerous.”
