Theoretically, taking rooibos with CYP2D6 substrates may increase the effects of CYP2D6 substrates.
In vitro research shows that the methanol extract of rooibos leaves and stems inhibits CYP2D6 enzyme activity.

This combination pill pairs aspirin with hydrocodone, an opioid pain reliever. Here's the wrinkle worth knowing: hydrocodone is actually a prodrug for part of its punch. Your body uses an enzyme called CYP2D6 to convert some hydrocodone into hydromorphone, a stronger painkiller. Rooibos has shown, in test-tube studies only, a tendency to slow that same enzyme down.

If that effect held true in people, you might convert a little less hydrocodone, so the pain relief could feel slightly weaker. Honestly though, this is based on lab data alone, not real patients, and a cup or two of rooibos tea is very unlikely to cause trouble. Concentrated extracts are a bigger question mark, so check with your pharmacist before using those.

Here's the gist with this combo pill. The hydrocodone part of your medication is processed in the body partly by a liver enzyme called CYP3A4. In a test tube, concentrated rooibos extract slowed that enzyme down. In theory, if rooibos blocks CYP3A4, your body could clear hydrocodone a bit slower, letting it build up. That might mean stronger effects like extra drowsiness, dizziness, nausea, constipation, or slower breathing.

But keep this in perspective. This is based only on lab studies, not real people, and a normal cup or two of rooibos tea is very unlikely to cause trouble. The bigger question is with concentrated rooibos supplements. If you use those regularly, just mention it to your pharmacist or doctor first.
