Why this applies to Maprotiline

Maprotiline falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES — the group 1,3-dmaa interacts with.

1,3-DMAA strongly inhibits cytochrome P450 2D6 (CYP2D6) enzymes in vitro. Theoretically, 1,3-DMAA can increase levels of CYP2D6 substrates. Some of drugs that are CYP2D6 substrates include amitriptyline (Elavil), clozapine (Clozaril), codeine, desipramine (Norpramin), donepezil (Aricept), fentanyl (Duragesic), flecainide (Tambocor), fluoxetine (Prozac), meperidine (Demerol), methadone (Dolophine), metoprolol (Lopressor, Toprol XL), olanzapine (Zyprexa), ondansetron (Zofran), tramadol (Ultram), trazodone (Desyrel), and others.

Here's the gist for maprotiline: your liver uses an enzyme called CYP2D6 to help break it down and clear it from your body. In lab studies, 1,3-DMAA appears to block that enzyme. If that happens in real life, maprotiline could build up higher than expected, which may amplify its usual side effects, things like drowsiness, dry mouth, constipation, dizziness, a fast heartbeat, or in rarer cases seizures. Maprotiline is not a prodrug, so more drug means stronger effects, not weaker.

On top of that, DMAA is a powerful stimulant that can spike your blood pressure and heart rate, which is risky to stack with this antidepressant. The enzyme data here is only test-tube level, so it's theoretical, but DMAA itself is banned and unsafe. Please skip it and check with your pharmacist or doctor.
