SEROTONERGIC DRUGS
Why this applies to Acetaminophen, Chlorpheniramine, Phenylpropanolamine
Acetaminophen, Chlorpheniramine, Phenylpropanolamine falls under SEROTONERGIC DRUGS — the group 5-htp interacts with.
5-HTP can increase serotonin levels and cause serotonergic effects. Theoretically, combining serotonergic drugs with 5-HTP might increase the risk of serotonergic side effects, including serotonin syndrome and cerebral vasoconstrictive disorders. However, serotonin syndrome with 5-HTP has not yet been reported in humans. Monitor patients for signs of serotonin syndrome and other serotonergic side effects if using 5-HTP with serotonergic drugs.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
This combo product has three pieces: acetaminophen for pain, chlorpheniramine (an antihistamine), and phenylpropanolamine (a decongestant). The part that overlaps with 5-HTP is its mild effect on serotonin. Since 5-HTP raises serotonin too, stacking them could, in theory, nudge serotonin activity higher than you'd want.
To be honest, this concern is mostly theoretical and based on how these ingredients work, not on real reported cases. Still, it's worth knowing the warning signs: agitation, a racing heart, sweating, shivering, or feeling jittery. The decongestant alone can already cause some of that. I'd just run it by your pharmacist or doctor before pairing them, especially if you take 5-HTP regularly rather than once in a while.

From our pharmacist’s overview on the Acetaminophen, Chlorpheniramine, Phenylpropanolamine page
“A stuffed-up head sends people to Sinulin or Conex (acetaminophen, chlorpheniramine, and phenylpropanolamine) without much thought, but this combination deserves genuine caution around supplements. Fourteen of its 292 known interactions are rated major, and they cluster in three ways: stimulant herbs such as ephedra can stack on phenyl…”
