5-htp and Desipramine

These two interact in 2 distinct ways. Each one is explained separately below.

Why Are There 2 Interactions?

5-htp interacts with whole categories of medication. Desipramine falls into 2 of those categories, so it can interact with 5-htp in 2 different ways. We’ve listed each one below — your overall risk is driven by the most serious of them (Moderate).

Moderate

CNS DEPRESSANTS

1 of 2

Why this applies to Desipramine

Desipramine falls under CNS DEPRESSANTS — the group 5-htp interacts with.

Theoretically, concomitant use of 5-HTP with medications that cause sedation might have additive effects.
In clinical trials, 5-HTP has been associated with drowsiness and somnolence.
Severity Moderate Likelihood Possible Evidence D

Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.

Dr. Brian Staiger explains PharmD, Clinical Pharmacist

Here's the simple version: both 5-HTP and desipramine can make you feel sleepy, so taking them together may add up to more drowsiness than either one alone. With desipramine you might already notice tiredness, a foggy head, or dizziness, and 5-HTP could nudge those feelings up a bit. Just so you know, this concern comes from how these two work on paper, not from strong studies, so it's more of a heads-up than an alarm.

There's a second piece worth a mention. Since 5-HTP raises serotonin and desipramine also affects serotonin, combining them could in theory pile on too much. I wouldn't start or stop anything on your own. Please run it past your pharmacist or doctor first.

SEROTONERGIC DRUGS

2 of 2

Desipramine also falls under SEROTONERGIC DRUGS.

Combining serotonergic drugs with 5-HTP might cause additive serotonergic effects.
5-HTP can increase serotonin levels and cause serotonergic effects. Theoretically, combining serotonergic drugs with 5-HTP might increase the risk of serotonergic side effects, including serotonin syndrome and cerebral vasoconstrictive disorders. However, serotonin syndrome with 5-HTP has not yet been reported in humans. Monitor patients for signs of serotonin syndrome and other serotonergic side effects if using 5-HTP with serotonergic drugs.
Severity Moderate Likelihood Possible Evidence D

Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.

Dr. Brian Staiger, PharmD, BCPS
Dr. Brian Staiger explains PharmD, Clinical Pharmacist

Here's the thing with desipramine: it's a tricyclic antidepressant that nudges serotonin (and norepinephrine) levels up in your brain. 5-HTP is a raw material your body turns into serotonin. So when you stack the two, you're pushing serotonin from two directions at once, and the worry is that it could add up.

This is a theoretical concern based on how both work, not something widely reported in real people, so I don't want to scare you. Still, it's worth watching for signs of too much serotonin: agitation, a racing heart, sweating, shivering, twitchy muscles, or confusion. If you notice those, get help. Before you combine these, please check with your pharmacist or doctor first.

5-HTP is a compound your body uses to make serotonin, and people take it as a supplement hoping to improve mood, sleep, and headaches.
